You have to use the right format, and the correct kind of transparency for that format.
PNG is one of the more common formats that permit transparency. In Photoshop, you need layers with transparent regions, which usually means getting rid of the special Background layer (which cannot be made transparent) or turning it into a regular layer (named "Layer 0" by default). You should see the checkerboard pattern that represents transparency showing through. It is better to use the "Save for Web" command, since you can verify that the transparency checkbox is enabled.
Certain other image formats—especially TIFF—can use an "alpha channel", which is a special channel (like the usual Red, Green, and Blue) that marks specific regions of the image as transparent, even if the layers themselves are fully opaque. When using the "Save As" dialog, make sure that the Transparency checkbox is enabled.
Be aware that PNGs cannot use Photoshop alpha channels, although the resulting images made with the method above actually do have an alpha channel.

Open the picture in photoshop.
Click and hold on the eraser, then choose magic eraser.
