Open screen shot in Preview

Use Select tool (from the Tools menu) and select Elliptical Selection

Drag Selection over area to be captured in the circle

Press Command C to copy the selected circle

Either Paste into where ever it is required or Select New from Clipboard in Preview's File menu and and save the circular image for later use.
