It is bit depending on your current background knowledge and future focus.

Say if you are already comfortable with C programming, then go for VC++ or Java

If your focus is towards VB or VB.NET then better plan accordingly.

Basically choosing a language for GUI development is based on the best fit for requirement. You can start with any one of those based on future aim.

You don't learn only GUI design...you learn that while learning software engineering since software engineering encompasses the user interface and its design. There really isn't a singular application that you learn to develop GUIs in...you can develop GUIs with Visual Studio, Xcode (actually called Interface Builder), QT Designer, and several others. Eclipse can be extended to work with various plug-ins that aid in "rapid" GUI design that you can then link code to (at least is the case with Android).

For GUI design you have two basic choices:

1] Flash (latest version) with flex. Flex is used for programming the behaviour of graphics in Flash.

2] JavaFX version 2. JavaFX 1 is obsolete while JavaFX 2 is popular new technology.

Latest version of Java provides support for JavaFX 2.

Moreover, you need to download specific plugin for Eclipse or NetBeans to use JavaFX 2.

JavaFX 2 is new while Flash and Flex are in use in most of the popular websites.

One more thing, Flash and Flex are for website designing only.

For stand alone applications you have only one option: JavaFX 2.

Moreover, basic applications can be developed even in Flash (with the help of coding in Flex)

but that will not help you to get a job. That is for entertainment purposes only.

Microsoft's user interfaces are moving towards being created in Expression. These tools allow you to completely separate program logic from the user interface and are beginning to get a great deal of industry traction.
